In addition, down regulation of 5-HTR2c has been reported in postmortem brains of patients with bipolar disorder and schizophrenia. It has also been shown that RNA editing of 5-HTR2c, a post transcriptional mechanism that deeply influences receptor function, is altered both in patients suffering of depression and in a rat model of the disease. The aim of this study was the evaluation of the possible relationship between 5-HTR2c and antidepressants treatment in the prefrontal frontal cortex and in the hippocampus of rats treated with fluoxetine and reboxetine. We analysed, in the rat P FC and in HI, the 5-HTR2c mRNA expression and the RNA editing level of the five editing sites, A, B, C, C' and D, present in the 5-HTR2c. Using a quantitative RT-PCR approach, we examined the relative changes in the expression pattern of the 5-HTR2c of rats chronically treated with reboxetine and fluoxetine 10 mg kg die ; . The expression pattern of 5-HTR2c was found altered by both the antidepressants. In particular we found, in P FC, a trend towards down-regulation that became significant -44% ; only after 3 weeks of treatment with both antidepressants. Otherwise, in HI, reboxetine induced a statistically significant alteration in 5-HT2c expression pattern only after three weeks of treatment, with a decrease of 56%, while fluoxetine induced a 2fold up-regulation at 2 and 3 weeks of treatment. In our protocol of treatment, the analysis of the RNA editing level at the 5 editing sites did not evidenced any biological alteration induced by the antidepressants. Moreover, the frequency of the different 5-HTR2c edited isoforms was determined by analyzing individual cDNA clones derived from RNA samples collected from the control group and from rats treated for 3 weeks with RBX and FLX both in HI and in P FC. We could not find significant alterations of RNA editing in any of the conditions examined. Our results suggest that chronic administration of antidepressants do not alter the editing level of 5HTR2c but have deep influence on its expression pattern in an area and time specific manner. Such modification of 5-HTR2c expression may contribute to the efficacy of SSRI NRI antidepressants.
