In the present study, evidence has been obtained that melatonin accelerates neuronal cell death in cultures. When added to native primary neuronal cultures, melatonin per se decreased cell viability as demonstrated by a significant increase in LDH within 72 h. This adverse effect of melatonin has not been documented before. Either no values for control cultures treated with melatonin were presented or the observation period was 24 h or shorter. At this time point, however, no change in cell viability had yet appeared in our experiments. No changes in LDH release within 72 h were detected in response to PBN and DMTU. The slight cytotoxicity of melatonin may indicate the presence of a prooxidant activity associated with the antioxidant melatonin. This phenomenon has been described for melatonin and other antioxidants including estrogen and vitamin C 57 60 ; The increase in malondialdehyde induced by melatonin also underlines the prooxidant potential of melatonin. According to Reiter 3 ; , melatonin is believed to work via electron donation in detoxifying the OH radical. In so doing, melatonin must itself become a radical, the indolyl cation radical. However, indole radical cations have been shown to react with oxygen, giving rise to indoxyls via peroxy radical intermediates. The formation of the radical cation of melatonin has been made responsible, e.g., for the enhancement of vitamin E consumption induced by oxidative stress in red blood cells 61 ; . Medina-Navarro et al. 60 ; also suggested the formation of secondary oxidation products as a possible mechanism for the prooxidant property of melatonin. A prooxidant action would also explain our findings that melatonin aggravated the AF64Aand staurosporine-induced LDH release, and the protective effect in OGD started to vanish in the higher dose range. Similarly, melatonin potentiated the neurotoxicity of methamphetamine in rats 62 ; . These findings, however, would implicate that caution is necessary, when melatonin is used therapeutically in higher concentrations in humans. To better understand METH effects in a more clinically relevant model, we studied the PK of METH and AMP in a self-administration paradigm. METHODS: Dual cannulated male rats n 5 ; were given METH via an infusion pump. The pump was programmed to deliver multiple METH injections of 0.048 mg kg based on actual METH selfadministration patterns in rats. Blood samples were collected at multiple time points and METH and AMP concentrations were determined by LC MS MS. RESULTS: Relatively stable METH concentrations were achieved within the first 30 min. of METH dosing. PK analysis showed that half-life, volume of distribution, total clearance and amount of AMP formed were not statistically different from values found after a single METH dose. CONCLUSION: These data showed rats self-administered loading doses of METH, which resulted in rapidly reaching steady-state concentrations. Our sample experienced significant decrements in Physical Functioning, Physical Role Limitations, Pain, General Health Perception, Energy, and Social Functioning, but not Emotional Wellbeing or Emotional Role Limitations. Comparing women with local recurrence LR ; , distant metastatic recurrence DR ; and NP disease, we found that DR patients reported a worse decline in general health perception compared to LR patients mean decline of 19.9 points vs. 6.4; p 0.001 ; . We identified important sources of response bias: DR women had higher death rates 28% vs 4% LR and 6% NP ; after agreeing to, but before completing the post-recurrence QOL assessment. The DR women were also more likely to refuse a post-recurrence QOL assessment 27% refusal rate vs. 14% LR and 13% NP ; , suggesting that patients with distant recurrence may be underrepresented in research examining the impact of recurrence on QOL. Federal amphetamine trafficking offenses, at this time, are very rare. Though the number of these cases is very small, the trends found in the Commission data track the trends in geographic diffusion and trafficker demographics observed in the mfthamphetamine data. The number of amphetamine trafficking cases received by the Commission from fiscal year 1993 through fiscal year 1998 has generally increased. The cases initially decrease from 27 cases in 1993 to 22 cases in 1994 and to a low of only 9 cases in 1995. However, the number of cases increase in 1996 to 36 and then to 131 cases in 1997. In 1998, 82 cases involved amphetamine as the primary drug. Similarities to methamphetaminf trends include the changing race and ethnicity of these offenders and diffusion pattern across the United States. The percentage of white offenders trafficking amphetamine has decreased steadily from 100 percent of the cases in 1993 to less than half by 1998 48.8 percent ; . There has been a corresponding increase in the percent of Hispanic offenders from zero percent in 1993 to 47.5 percent in 1998. Also, similar to recent trends in methamphetamine cases, the number of states in which an Hispanic offender was sentenced for the federal crime of amphetamine trafficking has been increasing from 0 in 1992 to 21 in 1998 ; . Demographic and Sentencing Summary Of the 82 amphetamine cases sentenced during 1998, offender and offense related information is available on 80 cases. Of these, approximately half are white 48.8% ; and half of Hispanic origin 47.5% ; . Males account for 83.8 percent of these offenders and 67.1 percent are United States citizens. Of these 80 offenders, 26 32.5 % ; received a departure for providing substantial assistance to authorities and an additional five cases 6.3% ; received a downward departure for other reasons. None received an upward departure. Seventy-three cases had information available on the quantity of amphetamine involved in the offense. Quantities ranged from 6.4 grams to 71, 820 grams. Half of these cases involved quantities greater than 500 grams and, thus, were eligible for a five-year sentence based on drug quantity alone. Of the 80 offenders described above, 78 received a sentence of imprisonment. The average sentence was 41 months.

He had a giant dental tumor that not only disfigured his face but threatened his life. People thought he was blind in the right eye; feeding and breathing had become a problem. Flown to Taiwan, a team of specialitsts removed the two pound tumor and reconstructed his face. Once shunned by his peers and neighbors, he was welcomed back by his community. A young man, remarkably handsome as a child developed a protruded jaw as he grew older. His bite being affected, he submitted to a maxillo-facial procedure that corrected the problem. The change in his looks was so dramatic, it took some time for his astonished family and friends to get used to his new face. He now intends to specialize in facial reconstruction after medical school. We have all witnessed the heartwarming smile of a baby, the twinkle in its parents' eyes as they watch it grow. We notice the permanent laugh lines around a grandmother's mouth, the set jaw of the determined athlete, the furrowed brow of a worried person. These facial clues give away the feelings of the person within and dictate how we interact with them. How many times have we looked at someone and made a quick judgment on whether they were likeable or not? Permanent unions have been forged from these first encounters. Later, they will insist that it was love at first sight. On the other hand, we have all been guilty of making adverse judgment and deciding we dislike someone until we get to know them better and change our impressions entirely. We are warned not to judge a book by its cover. It seems that faces are our fronts to the outside world, the covers that we want others to see. We are identified by them. To our knowledge, this investigation provides evidence for the first time that functional neuroimaging can be used to predict relapse in individuals with methamphetamine dependence. As hypothesized, methamphetaminedependent subjects who relapse during the follow-up interval show less activation in dorsolateral prefrontal, parietal, and temporal cortex, as well as insula, a network of structures that is critical for decision making.
