Estradiol

Estradiol induced similar relaxations in both sexes. Similarly, in rabbit coronary arteries, relaxation to estrogen 13 ; or raloxifene 9 ; showed no sex difference. Non-genomic effects of raloxifene may be mediated through activation of estrogen receptors. The selective estrogen receptor antagonist ICI 182, 780 did not antagonize raloxifeneinduced relaxation or raloxifene-induced inhibition of contraction and VSM [Ca2 + ]i increase in rings exposed to CaCl2 from either sex. In contrast, this blocker attenuated endotheliumdependent coronary artery relaxation to raloxifene 8 ; without affecting the endotheliumindependent effect 8, 22, 28 ; . ICI 182, 780 also inhibited raloxifene-stimulated NO formation in human endothelial cells 26 ; . Likewise, ICI 182, 780 antagonized only the endotheliumdependent relaxation to tamoxifen, another SERM 9 ; . Taken together, it appears that raloxifeneinduced endothelium-mediated effect involves ICI 182, 780-sensitive estrogen receptors; whereas the direct muscle relaxing action is probably unrelated to classic estrogen receptor stimulation. In conclusion, we have provided experimental evidence to show a principal mechanism by which raloxifene relaxes the rat intralobar renal arteries. Both raloxifene and 17-estradiol relax renal arteries through inhibiting Ca2 + entry mechanisms, probably via L-type Ca2 + channels. This effect is independent of a functional endothelium or ICI 182, 780-sensitive estrogen receptors, and it is equivalent in both sexes. Raloxifene induced significantly greater renovascular relaxation than 17-estradiol at submaximal concentration. Similar observations were also made in rabbit coronary arteries 8 ; . However, chronic and in vivo genomic effects of raloxifene may differ. Raloxifene is clinically used to treat menopausal women, but the present data show raloxifene is equally effective in causing renovascular relaxation in both sexes. However, chronic and genomic action of raloxifene on renovasculature may differ. Long-term oral intake of 60 mg day of raloxifene in women is expected to result in a mean maximal plasma concentration of 1.36 g L raloxifene 2.7 nM ; 14 ; . the present study, the minimal.
Objectives: To compare the sonographic endometrial growth pattern in conception and non-conception cycles during the luteal phase of ovarian stimulation cycles for in-vitro fertilization IVF ; and spontaneous cycles of frozen embryo transfer FET ; . Methods One hundred and sixty cycles of women undergoing assisted reproduction treatment were evaluated in the luteal phase. Endometrial data were related to the day of luteinizing hormone surge LH ; or the hCG administration day day 0 ; . Ultrasonographic evaluation of endometrial thickness was performed using 5MhZ transvaginal probe on day 0, day 4 and day 10. Serum oestradiol E2 ; and progestrone P ; levels were measured on the corresponding days. Results: In IVF n 100 ; , 23 conception cycles were compared with 87 non-conception cycle. There was no difference in the endometrial thickness, serum E2 and P levels between conception and non conception cycles at all time points in the luteal phase. In FET cycles n 60 ; , a similar trend was observed on day 0 and day 4, between conception n 13 ; and non-conception cycles n 57 ; . However, by the mid luteal phase the endometrium of conception cycles 122.3mm; mean SD ; demonstrated a significantly P 0.05 ; greater thickness than the non-conception cycles 10 2.9mm ; . Similarly, on day 10 serum E2 784422 pmol L ; and P levels 8330 nmol L ; in conception cycles were significantly higher than those in non-conception cycles E2 524434 pmol L., P 5452 nmol L ; . This pattern was not observed in IVF cycles. The sonographic thickness was similar at all time points in the luteal phase between conception and non-conception IVF cycles. Conclusions: The pattern of endometrial development in IVF cycles was different from that in FET cycles. These differences in the late luteal phase can be related to the endometrial growth pattern, serum E2 and P levels between conception and conception cycles. NORPACE . Disopyramide NORPLANT . S SELECTIVE AROMATASE INHIBITORS Needed were agents that inhibit aromatase-- and only aromatase. In theory, these would not cause the side effects associated with nonselective inhibitors such as aminoglutethimide, and, hopefully, could provide an effective treatment option. P450 inhibitors A group of azole compounds that reversibly inhibit fungal P450 enzymes also inhibit aromatase. Though these agents are intrinsically nonspecific and inhibit a variety of P450 subtypes, several have been developed that are highly specific for the aromatase P450 complex. This specificity should prevent any action against other P450 subtypes or interference with the synthesis of other steroid hormones. Two such agents that are available as oral preparations are anastrozole Arimidex ; and letrozole Femara ; .9 Androstenedione analogues Other compounds have structural similarities to androstenedione, the major substrate of aromatase. These agents irreversibly bind to aromatase, inactivating it permanently. Owing to their structural homology with androstenedione, they are highly specific for aromatase and do not alter the production of other steroid hormones.9 Exemestane Aromasin ; is the first aromatase inhibitor in this class available in the United States. This drug, given orally, has been shown to suppress estrone and estradiol levels by up to 95%.6 Owing to differences in mechanisms of action, exemestane may be effective in women in whom cancer has progressed on one of the reversible inhibitors, such as letrozole or anastrozole. s TRIALS OF SELECTIVE INHIBITORS IN METASTATIC BREAST CANCER Compared with aminoglutethimide These new, more-selective inhibitors of aromatase quickly made aminoglutethimide obsolete for treating breast cancer. Several trials demonstrated their superiority over aminoglutethimide, consistent with the obserVOLUME 69 NUMBER 7 J U.

Abstract #148 A Comparison Between Automated and Manual Blood Pressure Measurements in a Diabetes Outpatient Clinic Rashid O. Al Jawair, MD, and Ruth McManus, MD, FRCPC Objective: Automated blood pressure machines are increasingly being used by the public in nonmedical settings. Use of these machines should encourage patient participation in their hypertension care decisions. However, the accuracy of results is unknown. Therefore, we compared automatic and manual blood pressure readings in a diabetes clinic population and sought correlations between the results and patient demographics. Methods: Blood pressure measurements for 33 unselected patients 14 men and 19 women ; attending an outpatient diabetes clinic were obtained manually and using an automatic blood pressure machine similar to those available in commercial settings. The order of manual and automatic measurements was random. Differences between automatic and manual readings were calculated and correlated with sex, age, weight, body mass index BMI ; , and arm circumference immediately above the cuff site ; . Results are reported as mean SEM. Results: The patients had the following characteristics: age 50.4 2.9 years, weight 92.9 3.2 kg, BMI 32.6 1.1 kg m2, and arm circumference 32.3 0.8 cm. Automatic cuff systolic results were 141.1 3.2 mm Hg, and manual cuff systolic results were 132.1 3.4 mm Hg difference 9.0 1.7 mm Hg ; . Automatic cuff diastolic results were 76.8 2.2 mm Hg, and manual cuff diastolic results were 76.2 1.8 mm Hg difference 0.5 0.8 mm Hg ; . Diastolic pressures were more closely correlated R2 0.88, P 0.005 ; than were systolic pressures R2 0.74, P 0.005 ; . No significant correlations were found between blood pressure cuff differences and heart rate, age, weight, BMI, or arm circumference. Discussion: This prospective, nonrandomized study included 33 patients who presented to the adult endocrine clinic at St. Joseph's Health Centre in March 2005. Blood pressure measurements were taken during routine vital signs examination preclinically. Blood pressure was measured by automated sphygmomanometer Test Your Blood. Stop the Pain at the Source When it comes to alleviating migraines and other headaches, researchers say it's important that "withdrawal of abused medications and treatment of any subsequent withdrawal symptoms" be addressed Int J Clin Pract 2004; 58: 1142-51 ; . But how do you relieve migraine pain without drugs? The answer is to stop treating the symptoms and find the source. The Spine Source New research reveals that headache pain may also stem from problems in the spine. By correcting misalignments and dysfunctional areas in the spine -- known as vertebral subluxations -- doctors of chiropractic can restore movement to those areas causing the headache pain. And studies show chiropractic is highly effective in alleviating headaches. Chiropractic adjustments, or spinal manipulation therapy SMT ; , has "an effect comparable to commonly used first-line prophylactic [preventive] prescription medications for tension-type headache and migraine headache." J Manipulative Physiol Ther 2001; 24: 457-66. ; Research also shows that "both neck exercise low-intensity endurance training ; and spinal manipulation are effective in the short and long term when compared to no treatment. There is also evidence that spinal manipula. METHODS Both eyes of 79 normally menstruating women 37 migraine and 42 healthy control ; were included in the the study. Subjects underwent complete ocular examination, and SAP and SWAP analysis in both follicular 12th to 13th day of the cycle ; and luteal phases days 1 to 2 before bleeding ; of two consecutive menstrual cycles. Visual field analysis were performed using Humphrey Field Analyzer II Humphrey Instruments Inc, San Leandro, California ; with full-threshold, central 30-2 program. Visual fields were divided into four regions as superior temporal, inferior temporal, superior nasal and inferior nasal, respectively. Visual field mean sensitivity MS ; was calculated for all regions separately. RESULTS Five migrainous and eight control subjects were lost to follow-up. The mean age of migrainous n: 32 ; and control subjects n: 34 ; were 34.4 3.6 years and 35.1 3.3 years, respectively p 0.05 ; . Serum estradiol levels were significantly lower p 0.001 ; , and serum progesterone levels were significantly higher in luteal phase p 0.001 ; . Control subjects demonstrated decrease in MS values in luteal phase with SWAP tests p 0.04 ; . However, migraine patients demonstrated it with both SAP and SWAP tests. Regional MS values were not different in both phases for control group with both tests, while nasal field regions of migraine patients demonstrated lower MS values in luteal phase with SWAP tests p 0.01 ; . CONCLUSIONS The present study has provided further evidence for the existence of impact of the sex steroids on the visual field analysis of the migrainous patients. In addition to the assessment of intraocular pressure, menstrual cycle phases are important parameters to be considered in migrainous women at risk of glaucomatous optic neuropathy and finasteride.

Mechanisms, i.e. activation of RANKL or osteoblast transcription regulators such as Msx2 and Cbfa1 runx2 ; by which 17-estradiol or raloxifene regulate OPG in vascular smooth muscle cells, remain to be determined 31, 54 ; . In the present experiments, no significant differences were observed in the expression of MGP among 17-estradiol, raloxifene-treated or solvent-treated aortic smooth muscle cells. In all three groups, expression of MGP significantly increased throughout the duration of the experiment. This observation is in accordance with earlier reports 43 ; . Regulation of MGP by estrogen may be cell-specific as 17-estradiol upregulated MGP transcription within 48 hours of exposure in human breast cancer cells 51 ; . In addition, differences in regulation of MGP and OPG by estrogen and raloxifene are consistent with differences in endochondral processes characteristic of medial calcification observed in OPG and MGP knockout mice 31 ; . MCNULTY, Secretary Statement of Vice Chairperson Robert K. Bloom Before the Commission for consideration is a proposed rulemaking on Advanced Meter Deployment for Electricity Providers which proposes amendments to 52 Pa. Code Chapter 57. The purpose of this proposed rulemaking is to establish uniform procedures and standards for the deployment and customer selection of advanced meters and electric distribution company installation of advanced meter networks which may be required under the Electricity Generation Customer Choice and Competition Act ``Act'' ; . Comments from interested parties addressing the various proposals within these regulations are requested and are due within 30 days from the date this order is published in the Pennsylvania Bulletin. I encourage parties to comment on this proposal and specifically request that commentators consider my concerns with this rulemaking as delineated below. The issue of whether the function of metering should be unbundled and performed on a competitive basis is a threshold issue of the transition to a competitive electric generation market that has yet to be considered by this Commission. The Act neither specifies that metering should be unbundled or that it should remain a regulated distribution function. Section 2804 3 ; of the Act specifies that the Commission shall require unbundling to separate the charges for generation, transmission and distribution, and that ``The Commission may require the unbundling of other services.'' Pursuant to Section 2807 a ; of the Act, subject to Commission approval, the Electric Distribution Company ``EDC'' ; may require that the customer install, at the customer's expense, enhanced metering capability sufficient to match the energy delivered by the suppliers with consumption by the customer. The proposed Order states that the Commission concludes that metering should remain a regulated function of the EDC at this time, that the choice of generation supplier is inherently connected with the opportunity for customers and suppliers to choose any type of advanced metering equipment that is necessary to support new generation services, and that those consumers and suppliers choosing to use advanced metering equipment should pay for any incremental cost thereby incurred. This recommendation proposes that the EDC shall own and pay the acquisition cost of advanced meters, but sets up a tariffed surcharge mechanism which the EDC could and glucovance and estradiol, for example, estraiol side effects. 80% respond to therapy. ABSTRACT Acetylcholine, acting on presynaptic nicotinic receptors nAChRs ; , modulates the release of neurotransmitters in the brain. The rat dorsal raphe nucleus DR ; and the locus coeruleus LC ; receive cholinergic input and express the 7nAChR. In previous reports, we demonstrated that estradiol E ; administration stimulates DR serotonergic and LC noradrenergic function in the macaque. In addition, it has been reported that E induces the expression of the 7nAChR in rats. We questioned whether E increased the expression of the 7nAChR in the macaque DR and LC. We utilized double immunostaining to study the effect of a simulated preovulatory surge of E on the expression of the 7nAChR in the DR and the LC and to determine whether 7nAChR colocalizes with serotonin and tyrosine hydroxylase TH ; in macaques. There was no difference in the number of 7nAChR-positive neurons between ovariectomized OVX ; controls and OVX animals treated with a silastic capsule containing E Ecap ; . However, supplemental infusion of E for 5-30 hours to Ecap animals Ecap inf ; significantly increased the number of 7nAChRpositive neurons in DR and LC. In addition, supplemental E infusion significantly increased the number of neurons in which 7nAChR colocalized with serotonin and TH. These results constitute an important antecedent for study of the effects of nicotine and ovarian steroid hormones in the physiological functions regulated by the DR and the LC in women. J. Comp. Angus-sired heifers were allotted by age mean 4 mo ; , BW mean 135 kg ; , and sire n 4 ; to either a control uninfected, n 10 ; or infected group n 11 ; . Metacercariae of Fasciola hepatica were administered intraruminally, d 0 ; to study effects on interval to puberty, circulating ovarian steroids, serum liver enzymes and BW. Blood samples were collected bimonthly from d 0 to and biweekly from d 60 through 210 for analysis of serum estradiol 17 E2 ; and progesterone P4 ; concentrations by RIA. At 2-wk intervals, BW was recorded, a blood sample was obtained to quantify serum aspartate-aminotransferase AST ; and -glutamyltranspeptidase GGT ; and a fecal sample was collected to assess excretion of F. hepatica eggs. Puberty was defined by the occurrence of the first luteal phase serum P4 concentrations 1.0 ng ml for a minimum duration of 10 d ; univariate ANOVA using the RANDOM statement in PROC GLM was used to determine significant linear and curvilinear responses to treatment in prepubertal heifers from d 0 to 113 ; for BW, E2 , P4 , AST, and GGT. Treatment effects at d 113 were determined by one way ANOVA. F. hepatica eggs were detected in all infected heifers after day 92. Linear P 0.01 ; and curvilinear P 0.05 ; responses for AST and a linear P 0.05 ; response for GGT concentrations were detected over time in infected heifers. On d 113, mean GGT levels were higher P 0.01 ; in infected than in control heifers 116.4 31.2 vs 20.2 2.8 U L, respectively ; . Mean BW, serum AST, E2 , and P4 concentrations did not differ between treatment groups on d 113. By d 210, 60% six of 10 ; of heifers in the control group and 36% four of 11 ; of heifers in the infected group attained puberty. We conclude that F. hepatica infection induced elevated levels of serum enzymes which are indicative of liver damage, and there was a more persistent elevation in GGT than the elevation in AST levels. Experimental fascioliasis resulted in a lower percentage of heifers that reached puberty within 7 mo of infection as compared to control heifers. Key Words: Heifer, Fascioliasis, Puberty.

Cordyceps Capsule Cordyceps sinesis ; In TCM, the various actions ascribed to Cordyceps Capsule are lung and kidney nourishing, vital essence and vital energy tonifying, hemostatic, and phlegm resolving or mucolytic. It is used in general debility after sickness, and for elderly persons. Its superior therapeutic effects have been confirmed in many controlled, well-designed studies carried out by medical schools in China including Beijing, Shanghai, and Nanjing. It is virtually nontoxic. The effects of this herb in chronic viral hepatitis have been studied. The efficacy rate was reportedly above 80% in a 256 patient clinical study. Cordyceps sinesis can lower ALT, improve liver function, relieve liver related symptoms, and increase albumin. It has also been used for cirrhosis caused by chronic viral hepatitis. In the previously mentioned study, 17 out of 22 patients had increased albumin levels after three months of treatment. Twelve of 17 patients with ascites experienced complete resolution of ascites while the other five had a reduction in ascites.1 This herb is helpful for immunodeficiency caused by viral infection, chemotherapy, radiotherapy, major illness or surgery. Cordyceps sinesis is used to treat impotence, premature ejaculation, low libido, low sperm counts and or activity, irregular menstruation, and leukorrhea. This herb can facilitate bile secretion and help expel sandy gall stones. It can relax the sphincter of Oddi. It can abate biliary obstruction and pain. It prevents the precipitation of gallstone forming elements.
