It has been proposed that TASK-3 channels may have roles in the genesis and or development of certain malignant tumours, although their exact roles are still not understood. In this study immunocytochemistry was performed to investigate the TASK-3 expression pattern of three melanoma cell lines maintained in tissue culture. The identity of the cells was demonstrated by using melanoma-specific markers HMB45 and S100 protein ; . The validity of the present findings was confirmed by employing three different primary antibodies targeting different epitopes of the human TASK-3 channel and by applying preadsorption control experiments. All the findings described in this work were consistently reproducible with all three primary antibodies. All three melanoma cell lines demonstrated intense TASK-3-specific labelling, whose pattern and distribution showed time-dependent changes. The TASK-3 specific reaction was never confined to the surface membrane of the cells, but intense labelling could be demonstrated intracellularly as well. It was noted that the dividing, spherical cells exhibited an intense, homogenously distributed TASK-3-specific labelling in all three cell lines. The flat, multiprocessal cells, on the other hand, showed somewhat less powerful labelling, which mainly concentrated to the nuclear-perinuclear region of the cell as well as to the developing processes. Rather surprisingly, intense TASK-3-specific labelling could also be demonstrated within the nuclei of the melanoma cells in all three cell lines. The nuclear expression was the most prominent in the proliferating areas and it was very much reduced in the confluent regions of the tissue culture. When the cell lines were exposed to an antimytotic agent Mitomycin C ; for 48 hours, the proportion of the TASK-3-positive nuclei was markedly reduced, especially in the non-confluent areas. Our results suggest that the inhibition of the cell-division affects the expression pattern of the TASK-3 channels, whereas interfering with the function of the TASK-3 channels may alter the cell development, proliferation and survival. These findings imply that TASK-3 channels might be involved in the regulation of cell-division and cell proliferation. Visceral hypersensitivity has been identified as an important pathophysiologic mechanism in patients with gastroesophageal reflux GERD ; , not well controlled by usual therapy. To assess the visceral perception and its relationship to therapy, we used the water load test WLT ; in patients with GERD. Methods. A symptom visual analogue scale VAS, 0-10 cm ; and 5 min-WLT was used to evaluate symptom sensitivity heartburn, postprandial fullness, vomiting, early satiety, nausea, bloating, epigastric pain, belching, epigastric burning ; in 10 healthy controls HC, 5F 5M, mean age 42 + 6 yrs ; and 12 patients with GERD 8F 4M, mean age 40 + 5 yrs ; , before and after therapy esomeprazole, 40 mg day for four weeks ; . Results. Before therapy, HC drank more water than GERD patients 588 + 42 ml 497 + 38 ml ; , without significant differences between men and women. GERD patients showed basally a significant correlation between WLT and symptoms VAS with respect to heartburn p 0.05 ; , nausea p 0.02 ; and belching p 0.05 ; . After 4-weeks PPI-therapy 7 GERD patients had significant improvement of the symptom VAS p 0.001 ; and only a little increment of the volume of water ingested 501 + 11 ; . GERD patients did not refer significant amelioration of the symptom VAS, and in 4 of them a significant correlation p 0.05 ; with WLT and symptoms VAS for post-prandial fullness and nausea was shown. Conclusions. GERD patients display gastric hypersensitivity compared to healthy controls concerning the water load test. This test can be useful to discriminate the GERD-subgroups that potentially not improve symptoms after the first step therapy, possibly due to an overlap with functional dyspepsia. P5.
